Post-doctoral position « Role of cytoskeletal proteins in the trafficking and inflammation of F508delCFTR protein, responsible for Cystic Fibrosis »
The post-doctoral position is currently available for 24 months in the team Canalopathies: cystic fibrosis and other diseases, of Research Center “Croissance et Signalisation”, INSERM U845, Faculté Necker, Paris, France.

Mission:
Our research concerns the mechanisms involved in Cystic Fibrosis (CF), a lethal genetic diseases due to mutations in CFTR gene. The most frequent mutation, F508del, leads to protein degradation, abnormal trafficking preventing mutated protein to be properly addressed to plasma membrane. This is associated with an abnormal transepithelial transport in secretory epithelia and chronic inflammation. We have shown recently that F508delCFTR binds to keratin 8, a cytoskeletal protein, and that the newly formed complex prevents the delivery of F508delCFTR to the plasma membrane (Colas at al Human Mol Gen 2011).
The post-doctoral project lies in the field of interactomic studies aiming to investigate involvement of cytoskeletal proteins in the trafficking and function of wild type/F508del CFTR. This will help to develop new therapeutic targets for CF. The candidate will have to use pulmonary cell lines and to develop the primary culture of cells derived from nasal brushings and/or lungs of CF patients. The experiments on mice might be necessary. The project will require multidisciplinary approaches including electrophysiology, protein biochemistry, immunoblot, co-immunoprecipitation, immunocyochemistry, ELISA. Collaboration with mass spectrometry teams will be developed.
Profile:
Candidates for this project should have a solid background in cell culture, cellular, molecular biology (transfection, real time PCR, plasmid construction), and biochemistry (Western Blot, co-immunoprecipitation, immunohistochemistry). The basis in electrophysiology would be an advantage.
